ABSTRACT

The chloroplast genome sequence of Nitraria tangutorum, a desert shrub, was sequenced using high-throughput sequencing technology and analysed phylogenetically in the present study. The chloroplast genome is 159,414 bp in length, including a large single copy region of 87,924 bp and small single copy region of 18,318 bp, and a pair of inverted repeat regions of 26,586 bp. The chloroplast genome contains 110 unique genes, including 77 protein-coding genes, four ribosomal RNA genes, and 29 tRNA genes. Most of these genes are present as a single copy and in two or more copies 19 genes occurred. Seventeen genes have one intron, and clpP and ycf3 genes contain two introns. A total of 81 simple sequence repeats (SSRs) were identified, most of them were found to be mononucleotide repeats composed of A/T. In addition to SSRs, 66 repeats were identified, including 41 tandem repeats, 10 palindromic repeats, and 15 forward repeats. The phylogenetic analysis based on 54 protein-coding genes demonstrated a close relationship between N. tangutorum and other plant species in Sapindales. The complete chloroplast genome sequence of N. tangutorum will provide important data for further study of taxonomy and systematics of the genus Nitraria.

ABSTRACT

Objective: To study the chemical constituents from the leaves of Nitraria tangutorum. Methods: The chemical constituents were isolated by solvent extraction, repeated silica gel chromatography, and preparative-HPLC. Their structures were elucidated by physicochemical properties and 1H-NMR and 13C-NMR data. Results: Nine compounds were isolated from the ethanol extract of the leaves of N. tangutorum and their structures were identified as 4-oxo-β-ionol β-D-glycopyranoside (1), (6R,9R)-3- oxo-α-ionol β-D-glycopyranoside (2), blumenyl C β-D-glucopyranoside (3), blumenyl A β-D-glucopyranoside (4), syringaresinol- 4'- O-β-D-glucopyranoside (5), citrusin B (6), nikoenoside (7), 2-phenylethyl-β-D-glucopyranoside (8) and indole-3-ethyl-β- D-glucopyranoside (9). Conclusion: All the isolates are reported from the plants of Nitraria L. for the first time and the 13C-NMR data of compound 9 are also reported for the first time.

ABSTRACT

Aim To observe the cytotoxic and proliferous effects of Nitraria tangutorum flavone(NTF)on human A-704 strain cells. Methods In vitro cultured human A-704 strain cells, the cytotoxic and proliferous effects of NTF on human A-704 strain cells were determined with methods of trypan-blue and clone formation, and the microelement in cell was detected with X-electron microscope scanning. Results ① The trypan-blue detection showed that the living cells in the NTF group(0.1,0.5 and 5 mg?L-1)and the 5-FU group were decreased by 33.8%~57.3%,35.8%~80.1%,60.8%~99% and 40.3%~59.6% at 6~18 hour after treatment, which significantly differed from those of the control group(P
